As a provider of dedicated expert services around Induced Pluripotent Stem Cells in culture, Cellected aims to deliver high quality clonal cell lines as fast as possible to research scientists.
To achieve this, they have established a robust genomic QC based on a combination of traditional G-Banding karyotyping and Stem Genomics’ iCS-digital™ PSC assay.
Cellected offers bespoke services relating to iPSCs, including production of clonal iPSCs and cell line QC, cryopreservation and characterization. “As research scientists working on hPSCs, we knew before starting the company how important regular in-process genomic control was in order to deliver consistent quality in the final product,” says Dr. Claire Richards, CEO and Founder. “For this reason, we set up a robust QC combining traditional G-Banding karyotyping with the targeted digital PCR-based assay iCS-digital™ PSC”.
Cellected particularly likes the quick turnaround time of Stem Genomics’ digital PCR assay that enables them to scale up their iPSC culture, without compromising genomic stability.
A great example of this efficient in-process QC is shown below. Here are the results obtained at 3 different passages for the same line, monitoring 24 known hotspots for copy number changes/CNVs.
According to Claire, “we received these results in days, which enabled us to keep a close eye on our cultures for the appearance of any sub-clones with copy number changes. This allowed us to make a decision earlier as to which iPSC cell lines/clones were more stable and suitable for banking”.
“I would recommend it to anyone who wants a good way of keeping an eye on their cells almost in real time and as a complement to traditional techniques that take a lot longer”.
Cellected has extensive expertise in iPSCs – especially clonal cell line derivation, process and protocol development, QC, characterization and banking. They also have a semi-automated platform for clonal cell line derivation that allows the generation of high numbers of clonal cell lines, enabling the efficient generation of datasets for regulatory submissions.
